In 1706, following the death of the previous superior—Mother Élisabeth de Sainte-Anne Boulard de Denainvilliers—she was designated as successor. However, ecclesiastical and royal authorities forbade the holding of official elections; consequently, she assumed the role and served as the community's last prioress during its final years.
During her tenure, the monastery—a major stronghold of Jansenism—endured intense political and religious persecution at the hands of King Louis XIV and the Archbishopric of Paris. In 1707, the monarch confiscated the revenues of Port-Royal des Champs to transfer them to the Paris branch, thereby financially strangling the community.
The situation came to a head in October 1709, when the Council of State ordered the definitive expulsion of the nuns. Mother Louise de Sainte-Anastasie shared the fate of her fellow religious sisters; they were forcibly removed from the convent and dispersed among various
monasteries under surveillance, marking the formal dissolution and subsequent physical destruction of the abbey.
